Abstract
The researcher’s purpose was to understand, observe, predict and graph the enzymes function, activity, structure and concentration. The researcher hypothesized that enzyme concentration, temperature and pH will increase or decrease enzyme activity rate depending on intensity. In the first activity, the researchers placed enzyme solution, distilled water and starch solution into 3 different medicine cups. They used a spot plate to determine enzyme activity using glucose and starch tests, by placing a specific amount of drops into each well with either of the solutions or water. In activity 2, the researchers tested the effect of enzyme activity on diastase activity. They used the same pipettes and medicine filled cups to begin their procedures. The testers placed distilled water and enzyme solution into the 12 wells and then dropped one drop of starch indicator into the first well. After 30 seconds they dropped another drop into the next well and continued this step throughout the 12 wells with 30 seconds intervals. Activity 3, the researcher placed two drops of starch solution into the 4 corners of the spot plate and then added a drop of cold enzyme solution into the first well. In the second well, they placed room temperature enzyme solution; in the third well, body temperature enzyme solution; in the last well, they placed boiling enzyme solution. In Activity 4, they observed the effect on pH on enzyme activity by placing 3 drops of distilled water into 3 separate wells on the spot plate. Activity 5, the testers did the protease activity by digesting gelatin by papain.
